Principle of the Assay::The Human ADAMTS4 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human ADAMTS4 in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Human ADAMTS4 coated on a 96-well plate. Standards and samples are pipetted into the wells and ADAMTS4 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human ADAMTS4 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of ADAMTS4 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: ADAMTS4, A disintegrin and metalloproteinase with thrombospondin motifs 4 is an enzyme that in humans is encoded by the ADAMTS4 gene. ADAMTS4 is a member of the large ADAMTS family of zinc-dependent proteases. The human ADAMTS4 gene is mapped to chromosome 1 by somatic cell hybrid analysis. The enzyme encoded by this gene lacks a C-terminal TS motif. It is responsible for the degradation of aggrecan, a major proteoglycan of cartilage, and brevican, a brain-specific extracellular matrix protein. The cleavage of aggrecan and brevican suggests key roles of this enzyme in arthritic disease and in the central nervous system, potentially, in the progression of glioma
